- Knockdown of TAR RNA binding protein 2, TRBP2 was achieved by transfecting MCF7 with TRBP2 specific siRNAs (Silencer® select Product # S13790, S13792). Western blot analysis (Fig. a) was performed using whole cell extracts from the TRBP2 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with TRBP Polyclonal Antibody (Product # PA5-84053, 0.4 µg/mL) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to TRBP2. No decrease in signal was observed in the uncharacterized band at ~50 kDa (*).

- CRISPR-Cas9 mediated genome editing ofTRBP (as confirmed by next generation sequencing) was achieved by using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR920707_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of TRBP was performed by loading 30 µg of HeLa wild type (Lane 1), HeLa Cas9 (Lane 2) and HeLa Cas9 transduced with TRBP Lentiviral sgRNA (Lane 3) whole cell extracts. The samples were electrophoresed using NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with TRBP Polyclonal Antibody (Product # PA5-84053, 1:500 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000 dilution). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556) with the iBright™ FL1500 (Product # A44115). A reduced signal in sgRNA transduced cells using the LentiArray™ CRISPR product line confirms that antibody is specific toTRBP. Uncharacterized band was observed in all the samples at ~50 kDa.

- Western blot was performed using TRBP Polyclonal Antibody (Product # PA5-84053) and a 39 kDa band corresponding to TAR RNA-binding protein 2; TARBP2 was observed across across the panel tested. Whole cell extracts (30 µg lysate) of MCF7 (Lane 1), MCF 10A (Lane 2), A549 (Lane 3), Hep G2 (Lane 4), Mouse Lung (Lane 5), Rat Lung (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX), 10 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.4 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).An uncharacterized band was also seen at ~50 kDa in all the tested cell lines.
