- Western blot analysis of Calretinin was performed by loading 25 µg of U87-MG (lane 1), SK-N-MC (lane 2) and mouse brain (lane 3) cell lysates onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4ºC overnight. The membrane was probed with a Calretinin monoclonal antibody (Product # MA5-14540) at a dilution of 1:50 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Plus Western Blotting Substrate (Product # 32132). Results show a band at ~29 kDa.

- Knockdown of Calretinin was achieved by transfecting HT-29 with Calretinin specific siRNAs (Silencer® select Product # s224821, s2325). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the Calretinin knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with Calretinin Monoclonal Antibody (SP13) (Product # MA5-14540, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Calretinin.

- Western blot analysis of Calretinin was performed using whole cell extracts (30 µg lysate) of HEL 92.1.7 (Lane 1), A-431 (Lane 2), U-2 OS (Lane 3), U-87 MG (Lane 4), HeLa (Lane 5) and tissue extract (30 µg lysate) of Mouse Brain (Lane 6). The blots were probed with Anti-Calretinin Rabbit Monoclonal Antibody (Product # MA5-14540, 1:250 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). A 31 kDa band corresponding to Calretinin was observed across the cell lines and tissue tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
