- Western Blot was performed using Anti-CD133 Polyclonal Antibody (Product # PA5-38014) and a 133 kDa band corresponding to Prominin-1 was observed across cell lines and tissue tested, with low expression in HeLa which is reported in the literature. Additional multiple uncharacterized bands (*) were observed across the panel. Membrane enriched extracts (30 µg lysate) of Caco-2 (Lane 1), HT-29 (Lane 2), HeLa (Lane 3), and Mouse Kidney (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
