( a ) Cartoon summarizing regulatory analysis of Sp7 locus by chromatin immunoprecipitation of H3-acetyl-K14, Smad1, endogenous Grhl3, or transiently expressed V5-GRHL3 from chromatin of immortalized E13.5 limb bud cell cultures grown in presence of recombinant BMP2 or Noggin. ( b ) QPCR analysis revealed BMP-dependent enrichment of these proteins at a putative consensus-binding motif for Grhl3 located at a ( c ) highly conserved genomic region ~13 kb upstream of the Sp7 transcription start site.
