Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/mL, fixed with 2% paraformaldehyde and washed with PBS. Cells were blocked with a 2% solution of BSA-PBS for 30 min at room temperature and incubated with a HER-4 monoclonal antibody (Product # MA5-13016) at a dilution of 0.5 µg/test for 60 min at room temperature. Cells were then incubated for 40 min at room temperature in the dark using a Dylight 488-conjugated goat anti-mouse IgG (H+L) secondary antibody and re-suspended in PBS for FACS analysis.

- Cross-linking of [ 125 I]HB-EGF to cells expressing HER1, HER2, HER3 or HER4 alone. ( A ) The NIH 3T3 cell lines expressing each of the four EGF receptor subtypes were cross-linked with DSS in the presence of 10 ng/mL (4x10 5 c.p.m./mL) of [ 125 I]HB-EGF and lysed. The lysates of cells expressing HER1 (lane 1), HER2 (lane 2), HER3 (lane 3) and HER4 (lane 4) were immunoprecipitated with anti-HER1, anti-HER2, anti-HER3 and anti-HER4 antibodies, respectively, separated on a 6% SDS-PAGE gel and the radioactive complexes were visualized by autoradiography. ( B ) HER4 cells were cross-linked with 10 ng/mL of [ 125 I]HB-EGF in the absence of neutralizing antibody (lane 1), in the presence of 5 mug/mL of neutralizing monoclonal anti-HER1 (LA1) (lane 2) or in the presence of neutralizing monoclonal anti-HER4 (H4.72.8) (lane 3) antibodies. The cells were lysed and the [ 125 I]HB-EGF-HER complexes analyzed on a 6% SDS-PAGE gel.
