- Western blot analysis of SLC7A11 in human HeLa, Huvec, HCT-116 and A549 cells. Samples were incubated in SLC7A11 polyclonal antibody (Product # PA1-16775) using a dilution of 2 µg/mL followed by an anti-rabbit HRP secondary antibody. Separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. Detection: chemiluminescence.

- Western blot analysis of SLC7A11 in 0.03 mg/mL human brain frontal cortex membrane lysate and human brain lysates. Samples were incubated in SLC7A11 polyclonal antibody (Product # PA1-16775) using a dilution of 100 µg/mL. (1) ladder; (2) no lysate + xCT, 100 µg/mL; (3) human brain frontal cortex membrane lysate, no primary antibody; (4-7) human brain lysates, 0.03 mg/mL, xCT, 100 µg/mL.

- Western blot was performed using Anti-SLC7A11 Polyclonal Antibody (Product # PA1-16775) and a 55 kDa band corresponding to SLC7A11 was observed across the cell lines and tissue tested. An uncharacterized band of ~ 36 kDa was also observed in some of the cell lines. Whole cell extracts (30 µg lysate) of MDA-MB-231 (Lane1), MCF7 (Lane 2), Hep G2 (Lane 3), HeLa (Lane 4), HCT 116 (Lane 5), K-562 (Lane 6), U-87MG (Lane 7) and tissue extract (30 µg lysate) of Mouse Stomach (Lane 8) were electrophoresed using Novex® NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution), using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Flow cytometry of SLC7A11 in HeLa cells. Samples were incubated in SLC7A11 polyclonal antibody (Product # PA1-16775) using a dilution of 5 µg/mL for 30 minutes at room temperature. Antibody (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Both antibodies were conjugated to DyLight 488.
