- Description: The eBioHP40 antibody reacts with the p40 subunit of human IL-12 p70, and human IL-23. IL-12 is a 70 kDa heterodimeric cytokine composed of two covalently linked chains, p40 and p35, and is secreted primarily by activated monocytes, macrophages, and dendritic cells. IL-23 is a biologically-related, heterodimeric cytokine composed of disulfide-linked p40 and p19 subunits. The eBioHP40 antibody has slightly lower background staining than C8.6 and recognizes a different epitope. Applications Reported: This eBioHP40 (HP40, HP-40) antibody has been reported for use in intracellular staining followed by flow cytometric analysis. Applications Tested: This eBioHP40 (HP40, HP-40) antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of cultured human dendritic cells. This can be used at 5 µL (0.03 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. eFluor® 660 is a replacement for Alexa Fluor® 647. eFluor® 660 emits at 659 nm and is excited with the red laser (633 nm). Please make sure that your instrument is capable of detecting this fluorochome. Excitation: 633-647 nm; Emission: 668 nm; Laser: Red Laser. Filtration: 0.2 µm post-manufacturing filtered.

- Staining of unstimulated (left) or stimulated (right) cultured human dendritic cells with Anti-Human CD11b PE (Product # 12-0118-42) followed by fixation and permeabilization and subsequent intracellular staining with Anti-Human IL-12/IL-23 p40 eFluor® 660. Total viable cells as determined by Fixable Viability Dye eFluor® 450 (Product # 65-0863-14) were used for analysis.
