- Western blot was performed using Anti-PAK4 Monoclonal Antibody (OTI1C7) (Product # MA5-26859) and a 64kDa band corresponding to PAK4 was observed across the cell lines and tissues tested. For Fig. A, whole cell extracts (30 µg lysate) of NIH:OVCAR-3 (Lane 1), SK-O-V3 (Lane 2), HCT 116 (Lane 3), Colo 205 (Lane 4), T-47D (Lane 5), MDA-MB-231 (Lane 6), A549 (Lane 7), A-431 (Lane 8) and for Fig. B, tissue extracts (30 µg lysate) of Mouse Testis (Lane 1), Mouse Heart (Lane 2) and Rat Testis (Lane 3) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blots were probed with the primary antibody (1:2000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). As reported in literature, the expression is low in SK-OV-3 and Colo 205 compared to NIH:OVCAR-3 and HCT 116 respectively (DOI: 10.1073/pnas.0907481107, DOI: 10.1158/1541-7786.MCR-12-0466). Relative expression is also observed between Testis and Heart, a tissue that expresses low levels of PAK4.
