- Knockdown of GBA was achieved by transfecting NIH:OVCAR-3 with GBA specific siRNAs (Silencer® select Product # S501316, S534768). Western blot analysis (Fig. a) was performed using Whole cell extracts from the GBA knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with GBA Monoclonal Antibody (OTI1D12) (Product # MA5-26589, 1:2000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to GBA.

- Western blot was performed using Anti-GBA Monoclonal Antibody (OTI1D12) (Product # MA5-26589) and a 60 kDa band corresponding to GBA was observed across the cell lines tested. Whole cell extracts (30 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), HEK-293 (Lane 3), U-87 MG (Lane 4), NIH:OVCAR-3 (Lane 5) and LNCaP (Lane 6) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunohistochemistry was performed on paraffin-embedded carcinoma of human liver tissue. To expose target proteins, 10mM citric buffer, pH6.0, 120°C for 3min was used. Following antigen retrieval, tissues were probed with a GBA monoclonal antibody (Product # MA5-26589).
