- Western blot analysis of Rex1 was performed by loading 20 µg of MDCK whole cell lysate and 5uL of PageRuler Plus Prestained Protein Ladder (Product # 26619) per well onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane using the G2 Blotter (Product # 62288), and blocked with 5% milk in TBST for 1 hour at room temperature. Rex1 was detected at ~35 kD using a Rex1 recombinant rabbit monoclonal antibody (Product # 701159) at a concentration of 2 µg/mL in 5% milk in TBST overnight at 4C on a rocking platform, followed by a Goat anti-Rabbit IgG-HRP Superclonal™ secondary antibody (Product # A27036) at a concentration of 1 µg/mL for at least 30 minutes at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico substrate (Product # 34080).

- Western blot analysis of Rex1 in A431 whole cell extracts using a Rex1 recombinant rabbit monoclonal antibody (Product # 701159) at a dilution of 2 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~35kDa.

- Immunofluorescent analysis of Rex1 in HeLa cells using a Rex1 recombinant rabbit monoclonal antibody (Product # 701159) followed by detection using an Alexa Fluor 488-conjugated goat anti-rabbit secondary antibody (green) (Image A). Nuclei were stained using DAPI (Image B) and actin stained with Alexa Fluor 594 phalloidin (red) (image C). Image D is a composite image showing nuclear localization of Rex1.
