- Reconstitute with 0.2 mL of distilled water to yield a concentration of 500 µg/mL. Positive Control - WB: human Hela whole cell, human HEK293 whole cell, human K562 whole cell, human THP-1 whole cell, mouse spleen tissue, mouse thymus tissue. ICC/IF: A431 cell, NIH/3T3 cell. Flow: HL-60 cell, HEPA1-6 cell.|Store at -20°C for one year from date of receipt. After reconstitution, at 4°C for one month. It can also be aliquotted and stored frozen at -20°C for six months. Avoid repeated freeze-thaw cycles.

- Immunocytochemistry analysis of NUP214 in A431 cells. Antigen retrieval was performed with enzyme antigen retrieval (15 min). Samples were blocked with 10% goat serum and incubated in NUP214 polyclonal antibody (Product # PA5-114371) at a dilution of 2 µg/mL (overnight, 4°C), followed by DyLight 488 conjugated goat anti-rabbit IgG (30 min, 37°C) and DAPI at a dilution of 1:100.

- Immunocytochemistry analysis of NUP214 using anti-NUP214 antibody (Product # PA5-114371). NUP214 was detected in a section of NIH/3T3 cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum and then incubated with 2μg/mL rabbit anti-NUP214 antibody (Product # PA5-114371) overnight at 4°C. DyLight®594 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

The cells were blocked with 10% normal goat serum, and incubated with the primary antibody (1 μg/1x10^6 cells) for 30 min at 20°C. DyLight 488 conjugated goat anti-rabbit IgG (5-10 µg/1x10^6 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 µg/1x10^6) used under the same conditions. Unlabelled sample without incubation with primary antibody and secondary antibody (Red line) was used as a blank control.
