- Figure 2 ICP22 interacts with cellular transcription elongation factors. ( A ) HEK293 cells were transiently transfected with Flag-ICP22, either full length (FL) or the CCR. Cells were lysed, nuclear lysates were carefully separated for IP assays using control IgG or anti-Flag beads. Copurified proteins were determined by Western blot using the antibodies noted at the right; ( B ) His-ICP22 CCR was bacterially expressed and immobilized on Ni-NTA beads. GST-Cyclin T1 1-266 and GST-CDK9 FL were expressed separately in E.coli . Immobilized GST-Cyclin T1 1-266 and GST-CDK9 FL were eluted with reduced glutathione and then incubated individually with His-ICP22 CCR . Both input and proteins retained on the beads after washes were subjected to Western blot with GST and His-tag antibodies as noted; ( C ) Bar chart of the top ten gene ontology (GO) of biological processes set with a false discovery rate (FDR)
