Description

PA3-011 detects ERp29 protein in rat, mouse, hamster, canine, human, non-human primate, bovine and guinea pig samples. PA3-011 has successfully been used in immunofluorescence, immunoprecipitation and Western blot procedures. By Western blot, this antibody detects an ~29 kDa protein representing ERp29 in rat liver homogenate. Immunofluorescence data demonstrates that ERp29 is localized to the ER of rat thyrocytes using PA3-011. The PA3-011 immunogen was a 6-histidine epitope tagged full length recombinant ERp29 protein.

Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of MDCK (Lane 1), NIH/3T3 (Lane 2), Hep G2 (Lane 3), A-549 (Lane 4), MCF-7 (Lane 5), A-431 (Lane 6) and HEK-293 (Lane 7). The blot was probed with Anti-ERp29 Rabbit polyclonal Antibody (Product # PA3-011, 1 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). A 29 kDa band corresponding to ERp29 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard  (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
