Experimental details: Western Blot was performed using Anti-KIF11 Polyclonal Antibody (Product # PA3-16834) and a 120 kDa band corresponding to Kinesin-like protein KIF11 was observed across cell lines tested. Nuclear enriched extracts (30 µg lysate) of A-431 (Lane 1), HeLa (Lane 2), HEK-293 (Lane 3), Jurkat (Lane 4), FaDu (Lane 5), MDA-MB-231 (Lane 6), T-47D (Lane 7), U-2 OS (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:6000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Additional uncharacterized band was also observed at ~80 kDa.

Experimental details: Western blot analysis of KIF11 in 5 µg /lane cells transfected with the pCMV6-ENTRY control or pCMV6-ENTRY KIF11 cDNA. Samples were incubated in KIF11 polyclonal antibody (Product # PA3-16834). Cells were transfected with the pCMV6-ENTRY control or pCMV6-ENTRY KIF11 cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates were separated by SDS-PAGE and immunoblotted.

Experimental details: Knockdown of Kinesin-like protein KIF11 was achieved by transfecting HeLa with Kinesin-like protein KIF11 specific siRNAs (Silencer® select Product # s7904, s7903). Western Blot analysis (Fig. a) was performed using Nuclear enriched extracts from the Kinesin-like protein KIF11 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The Blot was probed with KIF11 Polyclonal Antibody (Product # PA3-16834, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:6000 dilution). Densitometric analysis of this western Blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Kinesin-like protein KIF11. Additional uncharacterized band was observed between 50-80 kDa.
