- Western blot was performed using Anti-14-3-3 beta protein (YWHAB) Polyclonal Antibody (Product # PA5-17425) on membrane enriched cell extracts (30 µg lysate) of A431 (Lane1), MCF7 (Lane 2), HEK-293 (Lane 3), Mouse brain (Lane 4), Mouse skeletal muscle (Lane 5), Rat brain (Lane 6) and Rat skeletal muscle (Lane 7) and 28 kDa band corresponding to 14-3-3 beta protein was observed in all samples except Mouse and Rat skeletal muscle. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 0.25 µg/mL, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
