- Knockout of CD26 was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR954602_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of CD26 was performed by loading 60 µg of Hep G2 wild type (Lane 1), Hep G2 wild type treated with 1X PTI for 4hrs (Lane 2), HepG2 Cas9 (Lane 3), HepG2 Cas9 treated with 1X PTI for 4hrs (Lane 4) , HepG2 CD26 KO (Lane 5) and HepG2 CD26 KO treated with 1X PTI for 4hrs (Lane 6) whole cell extracts. The samples were electrophoresed using NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with Anti-CD26 Recombinant Rabbit Monoclonal Antibody (JM11-42) (Product # MA5-32643, 1:500 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:8000 dilution) using the iBright™ FL 1500 (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556). Loss of signal upon CRISPR mediated knockout (KO) using the LentiArray™ CRISPR product line confirms that antibody is specific to CD26.

- Western Blot was performed using Anti-CD26 Recombinant Rabbit Monoclonal Antibody (JM11-42) (Product # MA5-32643) and a 120 kDa band corresponding to Dipeptidyl peptidase 4 was observed across all the tested cell lines and tissues, except Jurkat, Jurkat+ Protein Transport Inhibitor Cocktail, K-562 and Mouse Colon. Whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), Hep G2 (Protein Transport Inhibitor Cocktail (500X), 1X, 4 hours) (Lane 2), Caco-2 (Lane 3), Jurkat (Lane 4), Jurkat (Protein Transport Inhibitor Cocktail (500X), 1X, 4 hours) (Lane 5), K-562 (Lane 6), Mouse Thymus (Lane 7), Mouse Colon (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1/4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
