- Western Blot analysis of beta-2 Microglobulin using beta-2 Microglobulin Monoclonal Antibody (Product # MA5-36022) at a dilution of 1:500. Lane A: A431 whole cell lysate, Lane B: U937 whole cell lysate, Lane C: Raji whole cell lysate, Lane D: HeLa whole cell lysate, (Lysates/proteins at 30 µg per lane). The secondary antibody used was a Goat Anti-Mouse IgG (H+L)/HRP at 1:10,000 dilution. Developed using the ECL technique and performed under reducing conditions. Predicted band size: 14 kDa. Observed band size: 14 kDa.

- Knockout of B2M was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID 983707) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of B2M was performed by loading 30 µg of HeLa Wild Type (Lane 1), HeLa Cas9 (Lane 2) andHeLa B2M KO (Lane 3) membrane enriched extracts. The samples were electrophoresed using NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with Anti-beta-2 Microglobulin Monoclonal Antibody (34) (Product # MA5-36022, 1:500 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4,000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Loss of signal upon CRISPR mediated knockout (KO) using the LentiArray™ CRISPR product line confirms that antibody is specific to B2M.

- Immunocytochemistry-Immunofluorescence analysis of beta-2 Microglobulin in Hela cells. Cells were fixed with 4% PFA, permeabilized with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with beta-2 Microglobulin Monoclonal Antibody (Product # MA5-36022) (1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor 488-conjugated Goat Anti-mouse IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to Cytoplasm and cell membrane.

Cells were stained with beta-2 Microglobulin Monoclonal Antibody (Product # MA5-36022), then a PE-conjugated second step antibody.
