- IHC of ADPN-101AP on 20 µm thick frozen tissue sections cut using a cryomicrotome. Sections were fixed on gelatin coated slides suitable for immunostaining. Sections were permeabilized with 0.5% Triton-X100 in 10 mM PBS, pH 7.4 for 15 minutes. Blocked with 5X DiluOBuffer containing blocking proteins for 1 hour to reduce background and then incubated overnight with primary antibody ADPN-101AP (1:100 dilution in 1X DiluOBuffer). Following 3 washes with PBS, a biotinylated anti-rabbit IgG (1:200 dilution in 1X DiluOBuffer) was added as secondary antibody and incubated for 2 hours. Color was developed using streptavidin-conjugated HRP and ABC reagent kit from Vector. Slides were counter stained with methyl green, dehydrated gradually with increasing concentrations of ethanol and then with xylene. Slides were mounted using Paramount medium from Thermo Scientific. Samples are fixed in formalin. Figure A is negative control and Figures B and C are stained with ADPN-101AP.
