- Description: This 9E2 monoclonal antibody reacts with human CD335, which is also known as NKp46. Expressed on NK cells, CD335 is a member of the natural cytotoxicity receptor family that also includes NKp30 and NKp44. This type I transmembrane glycoprotein is involved in activating and expanding resting NK cells following interaction with dendritic cells to eliminate tumor and virus-infected (e.g., influenza) cells. CD335 also interacts with pancreatic beta cells during the development of type I diabetes. The 9E2 antibody has been reported to induce cytotoxicity. Applications Reported: This 9E2 antibody has been reported for use in flow cytometric analysis and to induce cytotoxicity in functional assays. Applications Tested: This 9E2 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 0.25 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Storage and handling: Use in a sterile environment. Filtration: 0.2 µm post-manufacturing filtered. Purity: Greater than 90%, as determined by SDS-PAGE. Endotoxin Level: Less than 0.001 ng/µg antibody, as determined by LAL assay. Aggregation: Less than 10%, as determined by HPLC.

- Normal human peripheral blood cells cells were stained with CD56 Monoclonal Antibody, APC (Product # 17-0566-42) and 0.125 µg of Mouse IgG1 kappa Isotype Control (Product # 14-4714-82) (left) or 0.125 µg of CD335 Monoclonal Antibody, Functional Grade (right) followed by Anti-IgG1 Monoclonal Antibody, Biotin (Product # 13-4015-82) and Streptavidin, PE (Product #12-4317-87). Cells in the lymphocyte gate were used for analysis.
