- Western blot analysis of ROR gamma in human and mouse Thymus, Jurkat and U2OS cells. Samples were incubated in ROR gamma polyclonal antibody (Product # PA5-23148) using a dilution of 2 µg/mL followed by an anti-rabbit HRP secondary antibody. Separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. Detection: chemiluminescence.

- Western blot was performed using Anti-ROR gamma Polyclonal Antibody (Product # PA5-23148) and a 55 kDa band corresponding to ROR gamma (t) was increased upon TGFB1 treatment. An uncharacterized band (*) at ~45 kDa was also observed. Modified whole cell extracts (1% SDS) (30 µg lysate) of DU 145 (Lane 1) and DU145 treated with TGFB1 (50 ng/ml for 48 hour) (Lane 2) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/ml) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunohistochemical analysis of ROR gamma in Tissue section of human lymph node. Samples were incubated in ROR gamma polyclonal antibody (Product # PA5-23148) followed by a HRP labeled secondary antibody and DAB reagent. Nuclei of cells were counter-stained with hematoxylin. This antibody generated mainly a nuclear staining in a subset of cells and a weak to cytoplasmic staining was also observed in some cells.
