Description
Naptumomab estafenatox (brand name Naptumomab) is an investigational immunocytokine fusion protein composed of a mouse monoclonal antibody fragment (Fab) targeting the tumor-associated antigen 5T4 (also known as TPBG) linked to a modified superantigen, staphylococcal enterotoxin E (SEE). The 5T4-specific Fab directs the construct to tumor cells expressing 5T4, while the SEE moiety acts as a polyclonal T-cell activator by binding to specific V¦Â regions of the T-cell receptor (TCR), causing massive local T-cell proliferation and cytokine release. This redirects pre-existing T cells to the tumor site, inducing an inflammatory anti-tumor response. It has been clinically evaluated for the treatment of advanced renal cell carcinoma (RCC) and non-small cell lung cancer (NSCLC).
