Detection of human and mouse WNK1 by western blot. Samples: Whole cell lysate (50 µg) from HEK293T, HeLa, Jurkat, TCMK-1, and NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-WNK1 antibody (Product # A301-515A; lot A301-515A-3) used for WB at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 3 minutes.

Experimental details

Detection of human WNK1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-WNK1 antibody A301-515A (lot A301-515A-2) used for IP at 6 µg per reaction. WNK1 was also immunoprecipitated by rabbit anti-WNK1 antibody A301-513A and A301-514A. For blotting immunoprecipitated WNK1, A301-515A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Antibodies: Affinity purified rabbit anti-WNK1 antibody (Product # A301-515A; lot A301-515A-3) used for IP at 6 µg per reaction. WNK1 was also immunoprecipitated by a previous lot of this antibody (lot A301-515A-2) and rabbit anti-WNK1 antibody (Product # A301-514A). For blotting immunoprecipitated WNK1, Product # A301-515A was used at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 75 seconds.
