- Adding 0.2 mL of distilled water will yield a concentration of 500 µg/mL. Positive Control - WB: human K562 whole cell, human PC-3 whole cell, human HepG2 whole cell, human HEK293 whole cell, human A549 whole cell, human U87 whole cell, rat liver tissue, rat kidney tissue, mouse kidney tissue. IHC: human adnexal serous adenocarcinoma tissue, human lung cancer tissue, human colorectal cancer tissue. Flow: U20S cell.|Store at -20°C for one year from date of receipt. After reconstitution, at 4°C for one month. It can also be aliquotted and stored frozen at -20°C for six months. Avoid repeated freeze-thaw cycles.

- Western blot analysis of LOC134147/CMBL using LOC134147/CMBL antibody (Product # PA5-143929). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50 µg of sample under reducing conditions. Lane 1: human K562 whole cell lysates; Lane 2: human PC-3 whole cell lysates; Lane 3: human HepG2 whole cell lysates; Lane 4: human HEK293 whole cell lysates; Lane 5: human A549 whole cell lysates; Lane 6: human U87 whole cell lysates; Lane 7: rat liver tissue lysates; Lane 8: rat kidney tissue lysates; Lane 9: mouse kidney tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-LOC134147/CMBL antigen affinity purified polyclonal antibody (Product # PA5-143929) at 0.25 μg/mL overnight at 4°C, then washed with TBS-0. 1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5,000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for LOC134147/CMBL at approximately 28KD. The expected band size for LOC134147/CMBL is at 28KD.

Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. Samples were incubated with CMBL Polyclonal antibody (Product # PA5-143929) using a dilution of 2 μg/mL overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

- Flow cytometry of CMBL in U20S cells (Blue line). The cells were blocked with 10% normal goat serum, and then incubated with CMBL polyclonal antibody (Product # PA5-143929) (1 µg/1x10^6 cells) for 30 min at 20°C. Isotype control antibody (Green line) was rabbit IgG (1 µg/1x10^6 cells) used under the same conditions. Unlabeled sample (Red line) was also used as a control. DyLight®488 conjugated goat anti-rabbit IgG (5-10 μg/1x10^6 cells) was used as secondary antibody for 30 minutes at 20°C.
