Experimental details: Immunofluorescent analysis of MCF-7 cells using a HRAS polyclonal antibody (Product # PA5-14863) at a dilution of 1:10-50, followed by a fluor-conjugated goat anti-rabbit secondary antibody (green). Actin filaments were stained with dye-conjugated phalloidin (red). Nuclei were stained with DAPI (blue).

Experimental details: Immunofluorescence analysis of HRAS was done on 70% confluent log phase A431 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were subsequently labeled with HRAS Rabbit Polyclonal Antibody (Product # PA5-14863) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:1500 for 45 minutes at room temperature. Nuclei (Blue) were stained with SlowFade® Gold Antifade Mountant DAPI (S36938). F-actin (Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). HRAS signal was observed in control cell line (panels a, d) and not in the HRAS knockout (KO) cell line (panels e, h). The images were captured at 60X magnification.

Experimental details: Immunocytochemistry analysis of H-Ras in MCF-7 cells. Samples were incubated in H-Ras polyclonal antibody (Product # PA5-14863) followed by Alexa Fluor 488-conjugated goat anti-rabbit lgG (green). Actin filaments have been labeled with Alexa Fluor 555 phalloidin (red). DAPI was used to stain the cell nuclear (blue).
