- Western blot analysis was performed on whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), HeLa (Lane 2), Jurkat (Lane 3), A549 (Lane 4), C2C12 (Lane 5), MOLT-4 (Lane 6), RAW 264.7 (Lane 7), U-87 MG (Lane 8), tissue extracts of Mouse Skeletal Muscle (Lane 9) and Mouse Liver (Lane 10). The blot was probed with Anti-TPI1 Polyclonal Antibody (Product # PA5-18342, 0.1 µg/ml) and detected by chemiluminescence using Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 0.25 µg/ml, 1:4000 dilution). A 31 kDa band corresponding to TPI1 was observed across cell lines and tissues tested.

- Knockdown of TPI1 was achieved by transfecting Hep G2 cells with TPI1 specific siRNAs (Silencer® select Product # s224747). Western blot analysis (Fig. a) was performed using whole cell extracts from the TPI1 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with TPI1 Polyclonal Antibody (Product # PA5-18342, 0.1µg/ml) and Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to TPI1.

- Immunofluorescence analysis of TPI1 was performed using 70% confluent log phase Hep G2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with TPI1 Polyclonal Antibody (Product # PA5-18342) at 1:100 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Rabbit anti-Goat IgG (H+L) Cross-Adsorbed Secondary Antibody, Alexa Fluor 488 (Product # A-11078) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytosolic localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

- Immunohistochemical analysis of TPI1 in paraffin embedded human liver using a TPI1 polyclonal antibody (Product #PA5-18342) at a concentration of 2 µg/mL. Steamed antigen retrieval was performed with pH 9 buffered Tris and EDTA. Samples were then stained with horseradish peroxidase.
