- Western blot was performed using Anti-BNIP3L Polyclonal Antibody (Product # PA5-29385) and 35-55 kDa bands corresponding to BNIP3L was observed across bands corresponding to multiple forms of BNIP3L were observed across cell lines tested and was upregulated upon cobalt chloride treatment in HeLa cells. Whole cell extracts (30 µg lysate) of HEL 92.1.7 (Lane 1), K-562 (Lane 2), HeLa (Lane 3), HeLa treated with cobalt chloride (15 µM for 48 hrs) (Lane 4), SK-BR-3 (Lane 5), HCT 116 (Lane 6) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
