- Immunoprecipitation was performed on HEK293T cells overexpressing FERMT2. Antigen-antibody complexes were formed by incubating the sample with 2 µg of FERMT2 monoclonal antibody (Product # MA5-24937) antibody and 0.1 mg of goat anti-mouse conjugated magnetic beads overnight. The sample was washed extensively and probed with rabbit anti-DDK polyclonal antibody.

- Knockdown of FERMT2 was achieved by transfecting MDA-MB-231 with FERMT2 specific siRNAs (Silencer® select Product # s21612, s21613 ). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the FERMT2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with Kindlin 2 Monoclonal Antibody (OTI9E4) (Product # MA5-24937, 1:1000 dilution) and Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to FERMT2.

- Western blot was performed using Anti-Kindlin 2 Monoclonal Antibody (OTI9E4) (Product # MA5-24937) and a 78 kDa band corresponding to Kindlin 2 was observed in A549, HT-1080, MDA-MB-231, BeWo cell lines; less in SK-BR-3 and none in K-562, which are known to be the cell lines that do not express Kindlin 2. Membrane enriched extracts (30 µg lysate) of A549 (Lane 1), HT-1080 (Lane 2), MDA-MB-231 (Lane 3), SK-BR-3 (Lane 4), BeWo (Lane 5), K-562 (Lane 6), tissue extracts of Mouse brown adipose (Lane 7) and Rat brain (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
