Western Blot of Rabbit anti-p27 antibody. Lane 1: HEK293 pcDNA3. Lane 2: HEK293 pcDNA3 Rb2wt. Lane 3: HEK293 pcDNA3 Rb2S941A. Lane 4: HEK293 pcDNA3 Rb2 PM19. Load: 30 µg per lane. Primary antibody: p27 antibody diluted in 1%BSA at 1:2000 for overnight at 4°C. Secondary antibody: IRDye800™ rabbit secondary antibody at 1:10,000 for 45 min at RT. Block: 5% BLOTTO overnight at 4°C. Predicted/Observed size: 27 kDa for p27.

Experimental details

Western blot using Rocklands affinity purified anti-p27 antibody shows detection of p27 protein in MCF7 whole cell lysate (lanes 1-3) (arrowhead). Separation was achieved using a 4-20% gradient gel. Blocking occurred using 5% BLOTTO. Primary antibody was diluted 1:500 in 1% BLOTTO. The membrane was washed and reacted with a 1:10,000 dilution of Dylight™ 800 conjugated Gt-a-Rabbit IgG. Molecular weight estimation was made by comparison to prestained MW markers indicated at the left (lane M). Other detection systems will yield similar results.
