- PA1-739 detects secretory carrier membrane protein 1 (SCAMP 1) from human, mouse and rat samples. PA1-739 has been successfully used in Western blot and immunoprecipitation procedures. By Western blot, this antibody detects an ~31 kDa protein representing SCAMP 1 in rat and mouse brain tissues as well as HeLa cell lysates. PA1-739 immunizing peptide corresponds to amino acid residues 1-17 from human SCAMP 1. This sequence is completely conserved between human, rat, and porcine SCAMP 1. PA1-739 immunizing peptide (Cat. # PEP-105) is available for use in neutralization and control experiments.

- Western blot analysis was performed on membrane extracts (30 µglysate) of U-87 MG (Lane 1), Neuro-2a (Lane 2), SH-SY5Y (Lane 3), SK-N-AS (Lane 4), T98G (Lane5), IMR-32 (Lane 6), and tissue extract (30 µglysate) of rat brain (Lane 7). The blots were probed with Rabbit Anti-SCAMP1 Polyclonal Antibody (Product # PA1-739, 2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 36 kDa band corresponding to SCAMP1 was observed across the cell lines and tissues tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
