- Detection of human Leo1 by western blot and immunoprecipitation. Samples: A. Whole cell lysate (WCL; 10, 30 or 50 µg) from control 293T cells (E) or 293T cells transfected with a Leo1 expression construct (T). B. Whole cell lysate (10mg) from 293T cells. Antibodies: Affinity purified rabbit anti-Leo1 antibody BL677 (Cat. No. A300-175A) used at 1 µg/ml for western blot in A and B. In B, Leo1 was immunoprecipitated by using BL677 at 3 µg/10mg with or without blocking peptide. Detection: Chemiluminescence with an exposure time of 1 minute. *The identity of the upper band is unknown but may represent a modified form of Leo1.

- Detection of human Leo1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-Leo1 antibody (Product # A300-175A; lot A300-175A-3) used for IP at 6 µg per reaction. Leo1 was also immunoprecipitated by a previous lot of this antibody (lot A300-175A-2). For blotting immunoprecipitated Leo1, A300-175A was used at 0.04 µg/mL. Detection: Chemiluminescence with an exposure time of 75 seconds.
