- The monoclonal antibody 11A8.7.4 reacts to DNAM-1 also known as CD226. DNAM 1 is a 65 kD receptor expressed by peripheral blood T lymphocytes that is involved in intercellular adhesion, lymphocyte signaling, cytotoxicity and lymphokine secretion mediated by cytotoxic T lymphocytes. DNAM 1 is broadly expressed on T cells, NK cells, platelets, monocytes and a subset of B cells. DNAM-1 is also expressed by a subset of CD3 positive thymocytes. The ligands for CD226 are the poliovirus receptor (PVR, CD155) and Poliovirus Related Receptor (PPR2, Nectin-2, CD112). CD226 is involved in cytolytic activity by interacting with integrins and activating protein tyrosine kinase Fyn. The 11A8.7.4 antibody has been shown to increase T and NK cell-mediated cytotoxicity. Applications Reported: This 11A8.7.4 antibody has been reported for use in flow cytometric analysis. Applications Tested: This 11A8.7.4 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
