- PA1-661 detects dynamin II (Dyn2) from rat, mouse, and human tissues and cells. PA1-661 has been successfully used in Western blot procedures. By Western blot, this antibody detects an ~100 kDa protein representing Dyn2 from HeLa cell lysate. PA1-661 immunizing peptide corresponds to amino acid residues 760-779 from human Dyn2. This sequence is 95% conserved in mouse and rat Dyn2. PA1-661 immunizing peptide (Cat. # PEP-056) is available for use in neutralization and control experiments.

- Immunofluorescence analysis of Dynamin II was performed using 70% confluent log phase HeLa cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Dynamin II Rabbit Polyclonal Antibody (Product # PA1-661) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoplasmic localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.

- Immunofluorescence analysis of Dynamin 2 was performed using 70% confluent log phase SH-SY5Y cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. - FIG 5 Cells depleted of AP-2 and dynamin II show reduced uptake of H. pylori vesicles. (A) H. pylori vesicles were incubated with siRNA-transfected AGS cells for 20 min. In comparison to vesicle uptake in AGS cells transfected with negative siRNA (100%), vesicle internalization was affected as follows in the knockdowns: AP-2, 63%; dynamin II, 80%; AP-2 plus dynamin II, 46%.
