- Knockdown of BID was achieved by transfecting Hep G2 cells with BID specific siRNAs (Silencer® select Product # s1986). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the BID knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with BID Monoclonal Antibody (Product # MA5-17034, 1:2000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to BID.

- Western blot analysis was performed on whole cell extracts of Hep G2 (Lane 1), Jurkat (Lane 2), MCF7 (Lane 3), PANC-1 (Lane 4), A-431 (Lane 5), HeLa (Lane 6), MOLT4 (Lane 7), SH-SY5Y (Lane 8) and PC-3 (Lane 9). The blot was probed with Anti-BID Monoclonal Antibody (3C5) (Product # MA5-17034, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/ml, 1:4000 dilution). A 20 kDa band corresponding to BID was observed across the cell lines tested.
