Experimental details: Knockdown of BID was achieved by transfecting Hep G2 cells with BID specific siRNAs (Silencer® select Product # s1986). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the BID knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with BID Monoclonal Antibody (Product # MA5-17034, 1:2000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to BID.

Experimental details: Western blot analysis was performed on whole cell extracts of Hep G2 (Lane 1), Jurkat (Lane 2), MCF7 (Lane 3), PANC-1 (Lane 4), A-431 (Lane 5), HeLa (Lane 6), MOLT4 (Lane 7), SH-SY5Y (Lane 8) and PC-3 (Lane 9). The blot was probed with Anti-BID Monoclonal Antibody (3C5) (Product # MA5-17034, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/ml, 1:4000 dilution). A 20 kDa band corresponding to BID was observed across the cell lines tested.
