- BID Recombinant Rabbit Monoclonal Antibody (JM11-14)
- Antibody type
- Monoclonal
- Antigen
- Synthetic peptide
- Description
- Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.

- Western blot was performed using Anti-BID Recombinant Rabbit Monoclonal Antibody (JM11-14) (Product # MA5-32642) and a 20 kDa band corresponding to BID was observed across cell lines and decreased upon Staurosporine and also with Actinomycin D along with TNF Alpha treatment. Whole cell extracts (30 µg lysate) of A-431 (Lane 1), A-431 treated with Staurosporine (1uM for 4 hr) (Lane 2), A-431 treated with Staurosporine (1uM for 8 hr) (Lane 3), A-431 treated with Actinomycin along with TNF Alpha (5ug/ml and 10ng/ml for 5 hr) (Lane 4), Hep G2 (Lane 5), Jurkat (Lane 6), MOLT-4 (Lane 7), HeLa (Lane 8), PC-3 (Lane 9) and SH-SY5Y (Lane 10) were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunofluorescence analysis of BID was performed using 70% confluent log phase A-431 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with BID Recombinant Rabbit Monoclonal Antibody (JM11-14) (Product # MA5-32642) at 5 µg/mL in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, Alexa Fluor 488 (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing cytoplasmic localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.
