- Western Blot was performed using Anti-RRM2 Polyclonal Antibody (Product # PA5-76703) and 44 kDa and 51 kDa band corresponding to Ribonucleoside-diphosphate reductase subunit M2 (RRM2) was observed across cell lines and tissues tested except in HT-29 and MCF7 which are reported low expressing for the protein. Whole cell extracts (40 µg lysate) of PANC-1 (Lane 1), HT-29 (Lane 2), MCF7 (Lane 3), MDA-MB-231 (Lane 4), HeLa (Lane 5), Mouse Thymus (Lane 6) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1:1500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
