- Western Blot was performed using Anti-NDUFB9 Polyclonal Antibody (Product # PA5-60110) and a 22 kDa band corresponding to NADH dehydrogenase [ubiquinone] 1 beta subcomplex subunit 9 was observed. Whole cell extracts (30 µg lysate) of SK-BR-3 (Lane 1), MCF7 (Lane 2), MDA-MB-231 (Lane 3), Hep G2 (Lane 4), Jurkat (Lane 5), A549 (Lane 6), COS-7 (Lane 7) and tissue extracts (30 µg lysate) of Mouse Spleen (Lane 8), Rat Spleen (Lane 9), Mouse Heart (Lane 10), Rat Heart (Lane 11) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.2 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Relative expression was observed between SK-BR-3, MCF7 and MDA-MB-231 as expected (https://doi.org/10.1371/journal.pone.0144441).
