Detection of human PGC-1beta by western blot. Samples: Whole cell lysate (50 µg) from HeLa, HEK293T, and MCF-7 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-PGC-1beta antibody (Product # A302-274A; lot A302-274A-2) used for WB at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 30 seconds.

Experimental details

Detection of human PGC-1beta by western blot and immunoprecipitation. Samples: Whole cell lysate (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-PGC-1beta antibody A302-274A used for WB at 0.4 µg/ml (A) and 1 µg/ml (B) and used for IP at 10 µg/mg lysate. PGC-1beta was also immunoprecipitated by rabbit anti-PGC-1beta antibody A302-273A, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 30 seconds (A and B).

Detection of human PGC-1beta by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PGC-1beta antibody (Product # A302-274A; lot A302-274A-2) used for IP at 6 µg per reaction. PGC-1beta was also immunoprecipitated by a previous lot of this antibody (lot A302-274A-1). For blotting immunoprecipitated PGC-1beta, Product # A302-274A was used at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 30 seconds.
