- Western blot analysis was performed on Nuclear extracts (30 µg lysate) of NIH/3T3 (Lane 1), 3T3-L1 (Lane 2), Hep G2 (Lane 3), COLO 205 (Lane 4), HeLa (Lane 5), HCT116 (Lane 6) and K-562 (Lane 7). The blot was probed with Anti-EHMT2 Monoclonal Antibody (B.133.9) (Product # MA5-14880, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 160 kDa band corresponding to EHMT2 was observed across the cell lines tested. An additional band around 60 kDa was observed in the cell lines tested.

- Knockdown of EHMT2 was achieved by transfecting HeLa cells with EHMT2 specific siRNAs (Silencer® select Product # s21468, s21469). Western blot analysis (Fig. a) was performed using whole cell extracts from the EHMT2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with EHMT2 Monoclonal Antibody (Product # MA5-14880, 1µg/ml) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/ml, 1:4000 dilution. Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to EHMT2.
