- ChIP assays were performed using NIH3T3 and HeLa cells, the anti-TIP5 crude serum (Product # 49-1037), and optimized PCR primer sets for qPCR (for the promoter and the coding region of the 28s ribosomal RNA gene). Each ChIP assay used 100 µg sheared chromatin (average fragment lengths between 200 and 400 bp, incubated overnight at 4°C) and either 5 µl crude serum against TIP5 or 5 µl IgG which was used as negative IP control. This figure shows the recovery by the TIP5 crude serum and by IgG (set to 1), normalized to the input DNA. These results show that, in both HEK293T and in NIH3T3 cells, TIP5 is associated with the promoter, but not with the coding region of the 28s rRNA gene.
