- Knockdown of HTR2B was achieved by transfecting SH-SY5Y with HTR2B specific siRNAs (Silencer® select Product # s7046, s7045). Western blot analysis (Fig. a) was performed using whole cell extracts from the HTR2B knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with HTR2B Polyclonal Antibody (Product # PA5-33304, 1:500 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to HTR2B.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), Hep G2 (Lane 2), MCF7 (Lane 3), T-47D (Lane 4), MDA-MB-231 (Lane 5), PC-3 (Lane 6), HEL 92.1.7 (Lane 7), tissue extracts of Mouse Brain (Lane 8) and Rat Brain (Lane 9). The blot was probed with Anti- HTR2B Polyclonal Antibody (Product # PA5-33304, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 54 kDa band corresponding to HTR2B was observed across all the cell lines and tissues tested.
