Description
Blosozumab is a humanized monoclonal antibody designed to target and inhibit sclerostin (SOST), a key negative regulator of bone formation. Sclerostin, primarily produced by osteocytes, acts by binding to low-density lipoprotein receptor-related proteins 5 and 6 (LRP5/6), thereby antagonizing the canonical Wnt/¦Â-catenin signaling pathway that is essential for osteoblast differentiation and activity. By neutralizing sclerostin, blosozumab aims to promote bone formation, increase bone mineral density (BMD), and enhance bone strength. It was investigated as a potential anabolic (bone-building) therapy for osteoporosis, demonstrating significant increases in BMD in Phase I and II clinical trials. However, its development for postmenopausal osteoporosis was discontinued in favor of another anti-sclerostin antibody, romosozumab, which later received regulatory approval.
