- HEK 293 cells stably transfected with Melanin-Concentrating Hormone Receptor 1 (MCHR1) were lysed in detergent buffer and glycoproteins were enriched using wheat germ lectin agarose beads. Receptors were eluted from the beads using SDS sample buffer for 20 min at 45 °C. Extracts were separated on 7.5% SDS-polyacrylamide gels and blotted onto PVDF membranes. Membranes were then incubated with Anti-MCHR1 Recombinant Rabbit Polyclonal Antibody (Product # 711649 at a concentration of 1 µg/mL). A broad band from 60-70 kDa corresponding to MCHR1 was observed.

- HEK 293 cells stably transfected with Melanin-Concentrating Hormone Receptor 1 (MCHR1) were grown on poly-L-lysine-coated coverslips overnight. After 24 h, cells were fixed with 4% paraformaldehyde and 0.2% picric acid in phosphate buffer (pH 6.9) for 30 min at room temperature and washed several times with phosphate buffer. Specimens were permeabilized with ice-cold methanol and then incubated with Anti-MCHR1 Recombinant Rabbit Polyclonal Antibody (Product # 711649 at a concentration of 1 µg/mL). Cells were then incubated with Alexa488-conjugated secondary antibody for 2 h at room temperature, mounted and examined using a laser scanning confocal microscope. Note, strong staining of transfected cells.
