- SH2D1A, also known as SAP, is a small adaptor protein which plays a major role in the bidirectional stimulation of T and B cells. SH2D1A is predominantly expressed in T cells and NK cells, and functions to regulate signal transduction pathways downstream of the SLAM family of surface receptors. The SH2D1A gene is frequently mutated in patients with X-linked lymphoproliferative disease. Malfunction of the SLAM-SAP pathway results in autoimmune diseases, tumors of the lymphoid tissues and inadequate responses to infectious agents.

- Total cell lysates (15 µg protein) from Jurkat (lane 1), mouse thymus (lane 2) and HeLa (lane 3, negative control) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with purified anti-SH2D1A (clone 1A9) antibody. Proteins were visualized using a goat anti-rat-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Purified anti-GAPDH (clone Poly6314) antibody was used as a loading control.
