- Western blot detection of BNIP3 in extracts of 293T cells transfected with human BNIP3. Samples were separated on SDS-PAGE and probed using a BNIP3 monoclonal antibody (Product # MA1-24688). The antibody was developed with AlkPh APA mouse Fab Ads Hu, Bov, Hor and an NBT/BCIP substrate.

- Western blot was performed using Anti-BNIP3 Monoclonal Antibody (ANa40)(Product # MA1-24688) and a 28kDa band corresponding to BNIP3 was observed across cell lines tested. Membrane enriched extracts (30ug µg lysate) of HeLa (Lane 1), HeLa treated with Cobalt Chloride, 150uM,48Hrs (Lane 2), MCF7 (Lane 3), MCF7 treated with Cobalt Chloride,150uM,48Hrs (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # LC2002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 Dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).Induced expression of BNIP3 was observed upon treatment with cobalt chloride in Hela and MCF-7 cells.
