Experimental details

Immunofluorescent analysis of LIN28B in HeLa cells. Samples were fixed with 4% paraformaldehyde (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with primary antibody (1:100, 2 h at room temperature). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 µg/mL, 5 min).

Experimental details

Flow cytometry analysis of LIN28B in HepG2 cells (green line). The cells were fixed with 2% paraformaldhyde (10 min) and permeabilized with 90% methanol (10 min). The sample was then incubated with 2% BSA followed by a LIN28B polyclonal antibody (Product # PA5-71920) using a dilution of 1:25 for 60 min at 37°C. The secondary antibody used was goat-anti-rabbit IgG, DyLight 488 with a dilution of 1:200 for 40 min at 37°C. Isotype control antibody (blue line) was rabbit IgG (1 µg/1x10^6 cells) used under the same conditions with an acquisition of >10, 000 events.
