- Western blot was performed using Anti-ROS1 Monoclonal Antibody (OTI1A1) (Product # MA5-26760) and a ~260kDa band corresponding to Proto-oncogene tyrosine-protein kinase ROS was observed across cell lines tested. Whole cell extracts (30 µg lysate) of MCF 10A (Lane 1), A549 (Lane 2), U-87 MG (Lane 3), HeLa (Lane 4), BeWo (Lane 5) and MCF7 (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005).

To expose target proteins, heat-induced epitope retrieval by 1mM EDTA in 10mM Tris buffer (pH8.0) at 120°C for 3 min. Following antigen retrieval, tissues were probed with a ROS1 monoclonal antibody (Product # MA5-26760) at a dilution of 0.2 µg/mL.
