- Polyclonal antibody for p27/CDKN1B detection. Host: Rabbit.Size: 100μg/vial. Tested applications: WB, IHC-P, IHC-F, ICC, FCM. Reactive species: Human. p27/CDKN1B information: Molecular Weight: 22073 MW; Subcellular Localization: Nucleus. Cytoplasm. Endosome . Nuclear and cytoplasmic in quiescent cells. AKT- or RSK- mediated phosphorylation on Thr-198, binds 14-3-3, translocates to the cytoplasm and promotes cell cycle progression. Mitogen- activated UHMK1 phosphorylation on Ser-10 also results in translocation to the cytoplasm and cell cycle progression. Phosphorylation on Ser-10 facilitates nuclear export. Translocates to the nucleus on phosphorylation of Tyr-88 and Tyr-89. Colocalizes at the endosome with SNX6; this leads to lysosomal degradation (By similarity); Tissue Specificity: Expressed in all tissues tested. Highest levels in skeletal muscle, lowest in liver and kidney.

- Western blot analysis of p27 KIP 1 using anti-p27 KIP 1 antibody (PB9070). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: Rat Thymus Tissue Lysate Lane 2: Rat Brain Tissue Lysate After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-p27 KIP 1 antigen affinity purified polyclonal antibody (Catalog # PB9070) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for p27 KIP 1 at approximately 27KD. The expected band size for p27 KIP 1 is at 27KD.
