- Knockdown of ATP Synthase B1 was achieved by transfecting HEK-293 cells with ATP Synthase B1 specific siRNAs (Silencer® select Product # s1777, s1779). Western blot analysis (Fig. a) was performed using membrane extracts from the ATP Synthase B1 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with ATP Synthase B1 Polyclonal Antibody (Product # PA5-27826, 1:500 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to ATP Synthase B1. A non specific band (*) was also observed at ~70KDa.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), HEK-293 (Lane 2), NIH/3T3 (Lane 3), Jurkat (Lane 4), Hep G2 (Lane 5), MOLT-4 (Lane 6) and tissue extract (30 µg lysate) of Mouse Brain (Lane 7). The blot was probed with Anti-ATP Synthase B1 Polyclonal Antibody (Product # PA5-27826, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 23 kDa band corresponding to ATP Synthase B1 was detected across the cell lines and tissue tested. An additional band were also observed at ~80kDa in certain cell lines.
