Description

PA1-076 detects GRASP55 from mouse and rat samples. PA1-076 has been successfully used in Western blot applications. By Western blot this antibody detects a ~55 kDa band representing the GRASP55 protein. The PA1-076 immunogen is a synthetic peptide corresponding to residues P(419) A S K V P T T V E D R V S D(433) of rat GRASP55 protein. This peptide is available for neutralization controls as PEP-295.

Western blot analysis was performed on membrane enriched extracts of HCT-116 (Lane 1) and HT-29 (Lane 2). The blot was probed with Anti-GRASP55 Rabbit Polyclonal Antibody (Product # PA1-076, 2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.4 µg/mL, 1:2500 dilution). A 55 kDa band corresponding to GRASP55 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
