- Western blot analysis of Fibrinogen alpha chain in human plasma (15 µg) . Samples were incubated with Fibrinogen alpha chain monoclonal antibody (Product # MA5-49571) with a dilution of 1 µg/mL, followed by peroxidase conjugated goat anti-rat lgG secondary antibody. FCA band was visualized using ECL Western Blotting Substrate. Result: MA5-49571 can detect human FGA by Western blotting.

- ELISA analysis of Fibrinogen alpha chain, microtiter wells were coated with Fibrinogen alpha chain Monoclonal Antibody (KT9) (Product # MA5-49571) at 5 µg/mL. Human FGA was used as the antigen. Peroxidase conjugated rat anti-human FGA monoclonal antibody (KT10) was used as the detection antibody Result: KT9 and KT10 can be used as a matched antibody pair to detect and quantify the concentration of FGA.

- Immunoprecipitation analysis of Fibrinogen alpha chain which was performed by incubating 2.5 µg/mL of Fibrinogen alpha chain in human plasma containing 200 µg total protein. After absorption with Protein G beads, the mixture was run on 6-18% SDS-PAGE and blotted onto nitrocellulose membrane. Samples were incubated with Fibrinogen alpha chain monoclonal antibody (Product # MA5-49571) with a dilution of 1 µg/mL, followed by peroxidase conjugated rabbit anti-mouse light chain specific lgG (which cross reacts with rat lgG) secondary antibody. Lane 1: Human plasma; Lane 2: Human FCA Immunoprecipitated from human plasma by Product # MA5-49571; Lane 3: The same as Lane 2 but with an lgG isotype control antibody. Result: MA5-49571 can immunoprecipitate human FCA.

- Immunohistochemistry analysis of Fibrinogen alpha chain in formalin-fixed paraffin-embedded human placenta tissue. Samples were incubated in Fibrinogen alpha chain monoclonal antibody (Product # MA5-49571) using a dilution of 2 µg/mL at room temperature for 1 hour followed by poly-peroxidase conjugated goat anti-mouse lgG (which cross reacts with rat lgG) secondary antibody. Antigen was retrieved thorough addition of boiling Tris/EDTA buffer pH 9 in a pressure cooker for 3 min. Endogenous peroxidase activity was quenched by incubating the sections with 3% H202 for 30 min at room temperature. Diaminobenzidine was used as the chromogen, and the section was counterstained with hematoxylin. Result: Decidual cells are positively stained at the cytoplasm and cell membrane.
